Catch a big bus/coach or minivan from Ekkamai bus station to Ban Phe pier, which cost roughly ฿120 and usually takes 3 hours. Ekkamai bus station is conveniently situated next to Ekkamai BTS station. I advise taking the big bus because it's comfier to sleep in as you can recline back and sleep, whereas in the minivan you can't. Once at Ban Phe Pier, you can take the ferry that is roughly another hour to reach Koh Samet.
