Tirumayam - Ramanathapuram National Highway 536, Kochi - Ernakulam - Munnar - Bodinayakkanur - Theni - Madurai - Sivaganga - Thondi National Highway 85, Vikravandi - Panruti - Neyveli - Kumbakonam - Thanjavur - Pattukottai - Manamadurai National Highway 36, Thanjavur - Pattukottai - Karaikkudi - Paramakudi - Sayalgudi State Highway 29, Ramanathapuram - Sivagangai - Melur State Highway 34, Nattarasankottai - Chokkanathapuram Major District Road 912, Okkur - Nattarasankottai Major District Road 300 are some of the major roads crossed by the Karaikkudi - Manamadurai Railway line.
