PCR stands for Polymerase Chain Reaction. PCR is a major molecular biological technique used to amplify DNA. Dr. Kary B Mullis, an American Biochemist invented PCR. Applications of PCR: In the diagnostic field, Biomedical Research, Forensic Sciences, Parenting disputes, etc. Types of PCR: Real-time quantitative PCR, Reverse transcriptase RT-PCR, Nested PCR, Hot start PCR, Multiplex PCR etc. Agarose gel Electrophoresis technique is used to visualize the amplified DNA in PCR. Reverse Transcription Polymerase Chain Reaction (RT-PCR) is used to detect and amplify Viral RNAs. RT-PCR is an accurate method than a rapid kit test for viruses. Requirements for PCR test: Template DNA, Primers, Taq Polymerase enzyme, Nucleotide bases, Buffer, and Thermal Cycler. PCR cycles: A typical PCR takes place in an automated thermal cycler machine consisting of a series of 20–40 repeated cycles with consistent temperature changes.
